Kothare started his career as a child artist with the film Maaja Chakula as Adinath, which earned him several accolades in 1994. He directed three follow-up films Chimni Pakhra (2003), Pachdalela (2004), and Khabardaar (2006). In 2008, he produced the film Full 3 Dhamaal. He appears in the 2010 film Ved Laagi Jeeva under the name Jaggu produced by his father Mahesh Kothare. He appeared in the 2011 film Stand By, playing the role of Rahul Narvekar. In the same year, he did a film Dubhaand in 2011 as Rohan Jadhav. Kothare played as Rego in 2012 in the musical drama Satrangi Re by Aditya Sarpotdar.
In 2013, she got engaged as Aditya Bolke in the first Marathi 3D film Japatlela 2. Kothare played the lead role of Nandan in the film Hello Nandan in 2014. He starred as Dr Vinay in the psychological thriller film Anvat in 2014. In the same year, he did the Marathi romantic drama film Ishq Wala Love as Ajinkya in 2014. He appeared in the lead role of Amod in the drama film Avatarachi Gosht in 2014.
Apart from films, Kothare made his debut as a producer with the television series Mann Udhan Variache in 2009. The series is a remake of the Bou Kotha Kao Bengali series.
